Plato-Bio: Verification-First Biological Novelty Screening with Temporal Rediscovery and Structural Benchmarks

Plato-Bio extends the open Plato/Denario research-agent architecture for biology by adding explicit workflow states, provenance records, citation checks, claim-to-evidence links, scoped file writes, and publication gates. After repairing three evaluation defects, the clean revision passed 931 Python tests, with six skips and no failures or errors. In one frozen historical rediscovery task, literature bridges ranked the later-studied fish-oil–Raynaud phenomenon relation first. In a separate benchmark comparing AlphaFold predictions with experimental structures for 15 human proteins, 11 high-confidence cores had C-alpha RMSD below 1 Angstrom, with a median of 0.501 Angstrom. The authors do not treat these results as evidence of prospective discovery.
